چکیده

We describe in this work the numerical treatment of the Filament-Based LamellipodiumModel (FBLM). This model is a two-phase two-dimensional continuum model, describing the dynamics of two interacting families of locally parallel F-actin filaments. It includes, among others, the bending stiffness of the filaments, adhesion to the substrate, and the cross-links connecting the two families. The numerical method proposed is a Finite Element Method (FEM) developed specifically for the needs of this problem. It is comprised of composite Lagrange–Hermite twodimensional elements defined over a two-dimensional space. We present some elements of the FEM and emphasize in the numerical treatment of the more complex terms. We also present novel numerical simulations and compare to in-vitro experiments of moving cells.

Stability, convergence, and sensitivity analysis of the Filament Based Lamellipodium Model and the corresponding FEM

This paper focuses on the study of the filament based lamellipodium model (FBLM) and the corresponding finite element method (FEM) from a numerical point of view. We study fundamental numerical properties of the FEM and justify the further use of the FBLM. We exhibit that the FEM satisfies a timestep stability condition that is consistent with the nature of the problem. We propose a particular ...

Mathematical modeling of Myosin induced bistability of Lamellipodial fragments

For various cell types and for lamellipodial fragments on flat surfaces, externally induced and spontaneous transitions between symmetric nonmoving states and polarized migration have been observed. This behavior is indicative of bistability of the cytoskeleton dynamics. In this work, the Filament Based Lamellipodium Model (FBLM), a two-dimensional, anisotropic, two-phase continuum model for th...

An extended Filament Based Lamellipodium Model produces various moving cell shapes in the presence of chemotactic signals.

The Filament Based Lamellipodium Model (FBLM) is a two-phase two-dimensional continuum model, describing the dynamics of two interacting families of locally parallel actin filaments (Oelz and Schmeiser, 2010b). It contains accounts of the filaments' bending stiffness, of adhesion to the substrate, and of cross-links connecting the two families. An extension of the model is presented with contri...

Role of actin-filament disassembly in lamellipodium protrusion in motile cells revealed using the drug jasplakinolide

BACKGROUND In motile cells, protrusion of the lamellipodium (a type of cell margin) requires assembly of actin monomers into actin filaments at the tip of the lamellipodium. The importance of actin-filament disassembly in this process is less well understood, and is assessed here using the actin drug jasplakinolide, which has two known activities - inhibition of filament disassembly and inducti...

Cofilin promotes stimulus-induced lamellipodium formation by generating an abundant supply of actin monomers

Cofilin stimulates actin filament disassembly and accelerates actin filament turnover. Cofilin is also involved in stimulus-induced actin filament assembly during lamellipodium formation. However, it is not clear whether this occurs by replenishing the actin monomer pool, through filament disassembly, or by creating free barbed ends, through its severing activity. Using photoactivatable Dronpa-...
